Pharmacological Mechanism

Cofactor for manganese-SOD (mitochondrial antioxidant), arginase (urea cycle), pyruvate carboxylase (gluconeogenesis), and glutamine synthetase. Neurotoxic at chronic excess — accumulates in basal ganglia, producing a parkinsonism-like syndrome (manganism).

• Iron (caution): Competitive DMT1 absorption — high Fe reduces Mn uptake and vice versa.
