3-Isobutyl-1-methylxanthine (IBMX) is a methylated xanthine derivative that acts as a non-selective competitive phosphodiesterase inhibitor (with IC50 values of 2 to 50 μM across most PDE families, excluding PDE8 and PDE9) and as a non-selective adenosine receptor antagonist.
In cell-based cAMP assays (such as HTRF, ELISA, or GloSensor), IBMX is added to the assay buffer alongside forskolin or GPCR agonists to block PDE-mediated hydrolysis so accumulated cAMP can be quantified at an endpoint.
Combined with insulin and dexamethasone in the classic "MDI" induction cocktail, IBMX elevates cAMP/PKA signaling to trigger C/EBPβ and PPARγ expression during in vitro differentiation of 3T3-L1 preadipocytes into mature adipocytes.
