PDE4A (phosphodiesterase 4A; HGNC:8780, NCBI Gene ID 5141) is a human locus mapped to chromosome band 19p13.2 that encodes cAMP-specific 3-prime,5-prime-cyclic phosphodiesterase 4A splice isoforms PDE4A1, PDE4A4/5, PDE4A10 & PDE4A11 (EC 3.1.4.53). PDE4A acts in compartmentalized hydrolysis of second messenger 3-prime,5-prime-cAMP to 5-prime-AMP -> termination of PKA/EPAC signaling, together with PDE4B (Schizophrenia & Neuroinflammation Paralog Gene), PDE4C (Ciliary & Pulmonary Paralog Gene), and PDE4D (Memory, Emesis & Brachydactyly Paralog Gene). Encodes cAMP-specific phosphodiesterase 4A, which compartmentalizes cyclic AMP degradation in prefrontal cortical dendritic spines, olfactory neurons, and inflammatory cells.
Compartmentalized hydrolysis of second messenger 3-prime,5-prime-cAMP to 5-prime-AMP -> termination of PKA/EPAC signaling
Clinical / Therapeutic Role
Prefrontal spine cAMP gating in working memory & sleep-deprivation cognitive deficits; co-targeted by roflumilast, apremilast & crisaborole
Gene Structure & Expression
Genomic Organization & Cytogenetic Locus: 17-exon gene on chromosome 19p13.2 (the human ortholog of the Drosophila learning-and-memory gene dunce), utilizing multiple alternative 5-prime exons to produce long isoforms containing both Upstream Conserved Regions (UCR1—bearing an activating PKA phosphorylation site—and UCR2) and short isoforms lacking UCR1, all sharing a C-terminal bimetallic (Zn2+/Mg2+) cAMP-specific catalytic domain.
Cellular & Tissue Expression Profile: Highly expressed in pyramidal neuron dendritic spines of the dorsolateral prefrontal cortex, hippocampal CA1 neurons, olfactory receptor neuron cilia (excluding the axonal knob), cerebral microvascular endothelium, and eosinophils/monocytes/T lymphocytes.
Molecular Function & Pathways
Biochemical & Physiological Mechanism: Anchors via isoform-specific N-terminal tails to scaffold proteins (such as AKAPs, DISC1, and p75NTR) to create nanometer-scale subcellular "cAMP sinks": by hydrolyzing the 3-prime-phosphoester bond of cyclic adenosine monophosphate (3-prime,5-prime-cAMP) into inactive 5-prime-AMP, PDE4A shapes local Protein Kinase A (PKA) and EPAC gradients; in prefrontal cortical dendritic spines, PDE4A is anchored near HCN cation channels by DISC1 so that basal cAMP is kept low, keeping HCN channels closed and strengthening working-memory network firing.
Clinical & Therapeutic Significance
Human Genetic Variants & Disease Phenotypes: Disruption of the DISC1-PDE4A/PDE4B complex or age-related loss of prefrontal PDE4A leads to excessive cAMP opening of spine HCN channels and working-memory impairment; conversely, acute sleep deprivation selectively up-regulates hippocampal PDE4A5 (PDE4A4 in humans), which degrades cAMP-PKA-CREB1 signaling and impairs hippocampal long-term potentiation and memory consolidation.
Therapeutic & Clinical Pharmacology: Co-inhibited across the catalytic pocket by FDA-approved pan-PDE4 inhibitors: oral roflumilast (Daliresp) and inhaled ensifentrine (Ohtuvayre, dual PDE3/PDE4) for COPD, oral apremilast (Otezla) for psoriasis and psoriatic arthritis, and topical boron-containing crisaborole (Eucrisa) and roflumilast for atopic dermatitis.
