Dominant-negative LoF causes Familial Partial Lipodystrophy Type 3 (FPLD3); Pro12Ala protects against T2D; target of pioglitazone & rosiglitazone
Gene Structure & Expression
Genomic Organization & Cytogenetic Locus: 9-exon gene spanning >140 kb on chromosome 3p25.2 driven by alternative promoters and 5-prime exon splicing that produce ubiquitous PPAR-gamma1 and adipose-restricted PPAR-gamma2 (which carries an additional 28 N-terminal amino acids conferring 5- to 10-fold higher ligand-independent AF-1 transcriptional activity).
Cellular & Tissue Expression Profile: PPAR-gamma2 is expressed almost exclusively in white and brown adipocytes, while PPAR-gamma1 is expressed in adipocytes, colonic epithelium, tissue macrophages (especially anti-inflammatory M2 and alveolar macrophages), vascular endothelium, and placental trophoblasts.
Molecular Function & Pathways
Biochemical & Physiological Mechanism: Acts as the indispensable master transcription factor of adipogenesis (no other factor can rescue adipocyte differentiation in Pparg-null cells): heterodimerized with RXRA and cooperating with C/EBP-alpha, PPARG activates FABP4 (aP2), LPL, CD36, GLUT4 (SLC2A4), and the insulin-sensitizing hormone adiponectin (ADIPOQ), safely sequestering circulating free fatty acids inside subcutaneous white adipocytes so liver and skeletal muscle are spared from lipotoxic insulin resistance; in brown/beige adipocytes, deacetylation of PPARG at Lys268/Lys293 by SIRT1 recruits PRDM16 and PGC-1alpha to induce UCP1.
Clinical & Therapeutic Significance
Human Genetic Variants & Disease Phenotypes: Heterozygous dominant-negative missense mutations in the ligand-binding domain (such as p.Pro467Leu and p.Val290Met) cause autosomal dominant Familial Partial Lipodystrophy Type 3 (FPLD3) with loss of limb/gluteal subcutaneous fat, severe insulin-resistant diabetes, hypertriglyceridemia, and hypertension; conversely, the common N-terminal polymorphism p.Pro12Ala (rs1801282) modestly tempers PPARG2 activity and reduces population risk of type 2 diabetes by ~15–20%.
Therapeutic & Clinical Pharmacology: Direct molecular target of thiazolidinedione (TZD) insulin sensitizers pioglitazone and rosiglitazone, which stabilize the AF-2 helix and block CDK5-mediated inhibitory phosphorylation of PPARG at Ser273 to treat type 2 diabetes and metabolic dysfunction-associated steatohepatitis (MASH).
