MYOD1 (myogenic differentiation 1; HGNC:7611, NCBI Gene ID 4654) is a human locus mapped to chromosome band 11p15.1 that encodes basic helix-loop-helix (bHLH) myogenic determination factor MyoD (320 aa). MYOD1 acts in skeletal myoblast lineage commitment, chromatin remodeling & transdifferentiation, together with MYOG (Myogenin Terminal Differentiation Gene), MYF5 (Myogenic Factor 5 Determination Gene), and MYF6 (Myogenic Factor 6 / MRF4 Maturation Gene). Encodes the pioneer basic helix-loop-helix transcription factor MyoD that commits mesodermal progenitors and satellite cells to the skeletal muscle lineage.
Genomic Organization & Cytogenetic Locus: 3-exon gene on chromosome 11p15.1 controlled by a core enhancer (-20 kb) and distal regulatory region (-5 kb), encoding a 320-amino-acid nuclear phosphoprotein containing an N-terminal transcriptional activation domain, a cysteine-histidine-rich chromatin-remodeling domain, and a basic helix-loop-helix (bHLH) domain carrying the conserved myogenic recognition motif Ala114-Thr115.
Cellular & Tissue Expression Profile: Expressed specifically in embryonic somitic myoblasts, activated adult skeletal muscle satellite stem cells following myofiber injury or resistance training, and mature fast-twitch glycolytic skeletal muscle fibers.
Molecular Function & Pathways
Biochemical & Physiological Mechanism: First single transcription factor shown to reprogram non-muscle fibroblasts, chondroblasts, and adipocytes directly into contractile skeletal myotubes (Harold Weintraub, 1987): MyoD heterodimerizes with ubiquitous E-proteins (E12/E47 encoded by TCF3) to bind canonical 5-prime-CANNTG-3-prime E-box motifs, recruits p300/CBP histone acetyltransferases and SWI/SNF (BAF) chromatin remodelers to open silent muscle loci, activates MYOG (myogenin) and sarcomeric structural genes, and induces p21 (CDKN1A) and RB1 to lock differentiating myocytes in permanent post-mitotic G0 arrest.
Clinical & Therapeutic Significance
Human Genetic Variants & Disease Phenotypes: Biallelic truncating loss-of-function mutations in human MYOD1 cause lethal congenital myopathy with fetal akinesia deformation sequence and diaphragmatic hypoplasia; conversely, a recurrent somatic heterozygous missense mutation in the DNA-binding domain (p.Leu122Arg, which converts MyoD into a MYC-like proliferation driver that fails to induce differentiation) defines aggressive spindle-cell and sclerosing rhabdomyosarcoma.
Therapeutic & Clinical Pharmacology: Repressed by Myostatin (MSTN)-SMAD2/3 signaling and inflammatory NF-kappaB/TNF in cachexia, whereas synthetic CRISPRa activation or lentiviral delivery of MYOD1 is widely used to transdifferentiate patient dermal fibroblasts into myotubes for diagnostic RNA splicing assays in Duchenne muscular dystrophy (DMD).
