CDKN1A (cyclin dependent kinase inhibitor 1A; HGNC:1784, NCBI Gene ID 1026) is a human locus mapped to chromosome band 6p21.2 that encodes intrinsically disordered nuclear/cytoplasmic CDK & PCNA inhibitor p21Cip1/Waf1 (164 aa; 21 kDa). CDKN1A acts in direct transcriptional effector of TP53 driving acute DNA-damage G1/S & G2/M cell-cycle arrest, PCNA replication block & senescence initiation, together with p21 (Cip1/Waf1) Protein Structure & Function, TP53 (Master Transcriptional Inducer of CDKN1A), and CDKN2A (p16INK4a Late Senescence Maintainer Gene). Encodes p21Cip1/Waf1, the direct transcriptional target of p53 that inhibits Cyclin-CDK2/CDK1 complexes and PCNA to halt DNA replication.
Genomic Organization & Cytogenetic Locus: 3-exon gene on chromosome 6p21.2 containing two high-affinity p53 (TP53) response elements at -2.3 kb and -1.4 kb in its promoter as well as SMAD4/TGF-beta, FOXO, VDR, and MYOD1 binding sites, encoding a 164-amino-acid Cip/Kip family polypeptide with an N-terminal Cyclin-CDK inhibitory domain and a C-terminal proliferating cell nuclear antigen (PCNA)-binding PIP-box motif.
Cellular & Tissue Expression Profile: Maintained at low basal levels in cycling cells, but rapidly induced within minutes of ionizing radiation, UV DNA damage, telomere uncapping, or replication stress via ATM/ATR-activated p53, and p53-independently induced during terminal differentiation of skeletal myoblasts (via MYOD1) and keratinocytes.
Molecular Function & Pathways
Biochemical & Physiological Mechanism: Stoichiometrically binds and inhibits Cyclin E/CDK2, Cyclin A/CDK2, and Cyclin B/CDK1 complexes to maintain RB1 in its hypophosphorylated, E2F-repressive state and halt G1-to-S and G2-to-M progression, while its C-terminal PIP-box binds trimeric sliding-clamp PCNA to competitively block DNA polymerase delta elongation without inhibiting nucleotide excision repair; at low stoichiometry, p21 also acts as an assembly factor for Cyclin D/CDK4 complexes.
Clinical & Therapeutic Significance
Human Genetic Variants & Disease Phenotypes: Unlike TP53 or CDKN2A, somatic coding mutations in CDKN1A are relatively infrequent (occurring primarily in ~10–15% of bladder urothelial carcinomas), because cytoplasmic AKT1-phosphorylated p21 (at Thr145) can paradoxically protect tumor and senescent cells from apoptosis by inhibiting procaspase-3 and ASK1.
Therapeutic & Clinical Pharmacology: In cellular senescence, CDKN1A (p21) drives the initial reversible DNA-damage arrest before CDKN2A (p16INK4a) locks in late irreversible senescence; selective targeting of p21-high senescent cells is an active frontier in fibrotic and metabolic disease research.
