MDM2 (MDM2 proto-oncogene; HGNC:6973, NCBI Gene ID 4193) is a human locus mapped to chromosome band 12q15 that encodes nuclear RING-finger E3 ubiquitin-protein ligase MDM2 / HDM2 (491 aa; EC 2.3.2.27). MDM2 acts in autoregulatory negative-feedback loop ubiquitylating & transcriptionally repressing p53 (TP53), together with TP53 (p53 Tumor Suppressor Degraded by MDM2), p53 Tumor Suppressor Protein, and CDKN2A (Encodes p14ARF Endogenous Inhibitor of MDM2). Encodes the RING-finger E3 ubiquitin ligase MDM2, the principal negative regulator that masks and degrades the p53 tumor suppressor.
Amplified in >90% of well-differentiated/dedifferentiated liposarcomas & sarcomas; target of MDM2-p53 antagonists (brigimadlin, navtemadlin)
Gene Structure & Expression
Genomic Organization & Cytogenetic Locus: 12-exon proto-oncogene on chromosome 12q15 driven by two promoters: a constitutive upstream P1 promoter and an internal p53-responsive P2 promoter in intron 1 (containing the functional SNP309 T>G polymorphism rs2279744 that creates a high-affinity SP1 binding site), encoding an N-terminal hydrophobic p53-binding pocket, a central acidic and zinc-finger domain (bound by p14ARF and ribosomal proteins RPL5/RPL11), and a C-terminal RING-finger E3 ubiquitin ligase domain.
Cellular & Tissue Expression Profile: Ubiquitously expressed in all tissues as part of a tight negative-feedback oscillator with p53, and massively overexpressed via 12q15 amplicons in soft-tissue sarcomas, osteosarcomas, and glioblastomas.
Molecular Function & Pathways
Biochemical & Physiological Mechanism: Restrains p53 through two concerted actions: its N-terminal hydrophobic cleft binds an amphipathic alpha-helix (Phe19, Trp23, Leu26) in the p53 transactivation domain to directly block p53 transcriptional activity, and its C-terminal RING domain (heterodimerized with MDMX / MDM4) polyubiquitylates p53 for 26S proteasomal degradation; when DNA damage activates ATM (which phosphorylates MDM2 at Ser395 and p53 at Ser15/Ser20) or oncogenic signaling induces p14ARF (CDKN2A), MDM2-p53 binding is disrupted and p53 accumulates.
Clinical & Therapeutic Significance
Human Genetic Variants & Disease Phenotypes: Homozygous deletion of Mdm2 is early embryonically lethal due to unchecked p53-driven apoptosis—a phenotype completely rescued by simultaneous deletion of Tp53; in human oncology, high-level focal amplification of MDM2 on ring or giant rod chromosomes is the pathognomonic driver of >90% of well-differentiated and dedifferentiated liposarcomas and intimal sarcomas (which retain wild-type TP53).
Therapeutic & Clinical Pharmacology: Structure-based small molecules (nutlins, idasanutlin, navtemadlin, milademetan, brigimadlin) and stapled alpha-helical peptides (sulanemadlin) mimic the Phe19-Trp23-Leu26 triad of p53 to occupy the MDM2 hydrophobic pocket and reactivate wild-type p53 apoptosis in MDM2-amplified sarcomas and hematologic malignancies.
