Lactone prodrug of simvastatin acid. Lipophilic — greater CNS penetration than rosuvastatin. CYP3A4 substrate — heavy interaction profile. Peak endogenous cholesterol synthesis occurs nocturnally → evening dosing for short-t½ statins.
