Convergence point of the two NAD⁺ biosynthesis routes. Produced either from nicotinic acid by NAPRT (Preiss-Handler salvage) or from quinolinic acid by QPRT (de novo tryptophan / kynurenine pathway). NaMN is then adenylylated by NMNAT1/2/3 to nicotinic acid adenine dinucleotide (NAAD), which is amidated by NADS to NAD⁺. Not a viable supplement — plasma nucleotide phosphatases and the absence of dedicated transporters prevent intact gut absorption. Precursor supplements (nicotinic acid, nicotinamide, NR, NMN) feed the same pathway at non-phosphorylated or monomeric-nucleotide entry points.
