Pharmacological Mechanism

Fully human IgG4 mAb binding IL-4Rα subunit shared by IL-4 + IL-13 receptors → blocks both Th2 cytokines. Paradigm-shifting treatment for atopic dermatitis — moderate-severe AD failed every prior systemic, dupilumab clears ~40% and improves 70–80%. Expanded to asthma, CRSwNP, EoE, prurigo nodularis.
