Core Principles & Analysis
Chemical calcium indicators such as fura-2, indo-1, fluo-3, and fluo-4 are derivatives of the chelator BAPTA (an EGTA homologue) selective for Ca²⁺ over Mg²⁺ and loaded into cells as membrane-permeable acetoxymethyl (AM) esters.
Genetically encoded calcium indicators (GECIs) such as GCaMP fuse green fluorescent protein (GFP), calmodulin, and the M13 peptide so Ca²⁺ binding induces a conformational change that brightens fluorescence.
In industrial pharmacology, high-speed calcium flux plate readers monitor Gq-coupled GPCRs and Ca²⁺-permeable channels (such as TRPV1 and NMDA receptors) across 384- and 1536-well microplates.
