Pharmacological Mechanism

Newer prodrug of tenofovir — preferential cleavage by lymphocyte cathepsin A → higher intracellular tenofovir-DP with much lower plasma tenofovir vs TDF. Reduced renal + bone toxicity. Replaces TDF in modern combination ART regimens.
