Reverses SERT, DAT, NET to cause non-exocytotic release of serotonin (principal), dopamine, noradrenaline. Also triggers oxytocin release. Subjective 'entactogenic' state (connection, empathy, emotional openness) distinct from classical psychedelics (5-HT2A agonism minor vs LSD/psilocybin). MAPS Phase 3 trials demonstrated strong PTSD efficacy; FDA rejected 2024 citing trial design concerns (second submission pending).
