Pharmacological Mechanism

Inactive prodrug — L-lysine conjugated to d-amphetamine. Cleaved by RBC peptidases to release active d-amphetamine. Enzymatic conversion is rate-limiting → flatter PK than immediate-release dextroamphetamine (lower Cmax, longer duration, less abuse potential when snorted/injected since conversion requires erythrocytes).
