Pharmacological Mechanism

Post-translational cleavage product of proglucagon, released from intestinal L cells in response to nutrient sensing. Activates GLP-1R on pancreatic β-cells (glucose-dependent insulin release), α-cells (glucagon suppression), hypothalamus (satiety), and gastric smooth muscle (slowed emptying). Rapidly inactivated by DPP-4 cleavage at the N-terminus — hence all clinical GLP-1 analogs carry Aib or other DPP-4-resistant substitutions.
