Pharmacological Mechanism

NRTI — guanosine analog, phosphorylated to carbovir-TP, a chain-terminator for HIV reverse transcriptase. HLA-B*5701 carriers (~5% of populations of European descent) develop life-threatening hypersensitivity → mandatory pre-treatment genotyping.
